Application | 3-chloropropyl methyl ether, also known as 3-methoxy-1-chloropropane, is an etherifying reagent that can be used to prepare antihypertensive drug aliskiren (Aliskiren) key intermediate 4-bromo-2-(3-methoxypropoxy)-anisole. |
synthesis method | add 90g(1mol)3-methoxy -1-propanol, 5g tetramethylammonium chloride and 200ml cyclohexane to a 500ml four-mouth bottle equipped with a thermometer and a dropping funnel; Maintain the reaction temperature at 65~70 ℃, stir and slowly drop 130.9g (1.1mol) of thionyl chloride, and finish dropping for about 3 hours, after the waste acid tail gas is discharged, alkaline water is absorbed, and then kept warm and stirred for 2 hours after dropping. The organic phase was washed with 100ml of water, 50ml of saturated sodium bicarbonate and 50ml of water respectively. After the solvent cyclohexane was recovered under reduced pressure, 106g of 3-methoxy -1-chloropropane was obtained by distillation and collection at bp103 ~ 106 ℃. The content of the product was 99% by gas chromatography and the yield was 97.7%. |
